Directed differentiation of iPSC


    Although iPSC has the potential to be re-differentiated into all kinds of somatic cells, increasing the efficiency and purity of re-differentiated cells has been a big challenge. Through more than five years effort, we have developed a robust and highly efficient process to make neural stem cells (NSCs) and neuronal progenitor cells (NPCs) from iPSC. We have consistently obtained high purity NSCs and NPCs from different iPSC lines with NSCs purity >95% and NPCs purity>90%. The whole process has been streamlined and adopted to large scale production. This core competency place CIB in a leadership position in China for NSC and NPC based cell therapy. Besides the neural lineage, we have also developed protocols for making cardiomyocytes, hepatocytes, and insulin-secreting beta-islet cells from iPSCs. We are currently optimizing protocols for those cell lineages and developing scale up manufacturing processes.

 

Neural cells derived from CIB iPSC lines

1. Neural stem cells (NSCs) derived from control (normal) iPSC line:

    This product is made from a fully characterized iPSC line derived from a normal healthy donor. The product comes as a kit containing one million NSC cells and culture media for NSC expansion. NSCs provided in this kit can be propagated at least three passages following our technical instruction. Expanded NSCs can be used directly for your experiment or be differentiated into neurons, astrocytes, and oligodendrocytes using commonly used protocols.

 

2. Neural stem cells (NSCs) derived from Park 7 knockout iPSC line:

    This product is made from an iPSC line with Park7 gene knockout.Park7 is a gene associated with Parkinson Disease. Dysfunction of Park 7 can result in defect of mitochondrial function and thus make dopaminergic neurons vulnerable to oxidative stress. Again, this product comes with one million NSC cells and expansion media. NSCs in the kit can be differentiated into mixed neurons and dopaminergic neurons.  

 

3.  Neural stem cells (NSCs) derived from Pink1 knockout iPSC line:

    This product is made from an iPSC line with Pink1 gene knockout. Like Park7, Pink1 is also a gene associated with Parkinson Disease. Dysfunction of Pink1 can result in defect of mitochondrial function and thus make dopaminergic neurons vulnerable to oxidative stress. This product comes with one million NSC cells and expansion media. NSCs in the kit can be differentiated into mixed neurons and dopaminergic neurons.  

 

4. Neuronal progenitor cells (NPCs) derived from control (normal) iPSC line:

    This product consists of neuronal precursor cells (NPCs) made using the NSC kit from a normal iPSC donor. The product comes as a kit containing one million NPCs and neuron maturation media. Following our technical instruction, one can obtain mature functional neurons in about one week and maintain the mature neuronal cells for about two to three weeks in vitro. This product can be directly used for neuron toxicity testing and for screening of neuron-protecting agents. In addition, this product can be used in basic research to study physiology of neurons.

 

5. Neuronal progenitor cells (NPCs) derived from Park7 knockout iPSC line:

    This product consists of neuronal precursor cells (NPCs) made using the Park7 knockout iPSC line. The product comes as a kit containing one million NPCs and neuron maturation media. Following our technical instruction, one can obtain mature functional neurons in about one week and maintain the mature neuronal cells for about two to three weeks in vitro. Neurons produced by this kit are more sensitive to neurotoxins and thus is good for testing chemical agents that can cause Parkinson disease and also for screening neuron-protecting agents.  This product is also useful for study the global effect of Park7 gene during neuron development.   

 

6. Neuronal progenitor cells (NPCs) derived from Pink1 knockout iPSC line:

    This product consists of neuronal precursor cells (NPCs) made using the Pink1 knockout iPSC line. The product comes as a kit containing one million NPCs and neuron maturation media. Following our technical instruction, one can obtain mature functional neurons in about one week and maintain the mature neuronal cells for about two to three weeks in vitro. Like the Park7 knockout neurons,Pink1 knockout neurons are more sensitive to neurotoxins and thus is good for testing chemical agents that can cause Parkinson disease and also for screening neuron-protecting agents.  This product is also useful for study the global effect of Pink1 gene during neuron development.
